After growing transformed E coli cells from the agar stab we received, we induced this part with IPTG in LB+amp and compared it to a positive control, p70417 in 1000 uM rhamnose, and to a negative control, p70417 in 0.2% glucose. We found that our part expressed higher GFP levels than the negative control, yet much lower than the positive control.j Note: expressed higher GFP levels than our other construct, LacDel.

Interpretation: Our construct showed fluorescence, but the dose-dependent curves are relatively flat. Therefore, while we seem to have eliminated the glucose inhibition, we seem to have also eliminated the lactose regulation of the lac operon, resulting in a constitutive promoter.
